Characterization of red wine native colloids by asymmetrical flow field-flow fractionation with online multidetection

Abstract Colloids are an important component of wines, but their study is challenging due to instability. Asymmetrical Flow-Field Flow Fractionation (AF4)-multidetection here proposed as a suitable approach isolate and characterize red wine colloids in native state. AF4 provided size-separation enabled quantification the colloidal content two wines. The gyration radius was determined by multi-angle light scattering, ranged between 25 50 nm. Analysis collected AF4-fractions showed that proteins, polysaccharides phenolics were present different proportions among fractions. composition differed SDS-PAGE analysis indicated presence protein-phenolics sub-aggregates only fractions containing with small radius. results allowed proposing model for structure, which comprises coexisting entities, one made covalently linked proteins-phenolics interacting non-covalent forces polysaccharides, second present. consistent reported relative stability fact can be stabilizing activity polysaccharides. Given enological practices affect extraction phenolics, AF4-technique represents very promising tool investigate effects these on aggregation behavior.
